Substitution of the poly-Pro II (φ,ψ) dihedral angles into this equation yields almost exactly Ω = -120°, i.e., the PPII helix is a left-handed helix (since Ω is negative) with three residues per turn (360°/120° = 3). == Properties of the imidazole side chain == At neutral or physiological pH, the imidazole side chain is neutral. The imidazole side chain in histidine has a pKa of approximately 6.0. Thus, below a pH of 6, the imidazole ring is mostly protonated and carries a positive +1 charge (as described by the Henderson–Hasselbalch equation). The resulting imidazolium ring bears two NH bonds and has a positive charge. The positive charge is equally distributed between both nitrogen atoms and can be represented with two equally important resonance structures. Sometimes, the symbol Hip is used for this protonated form instead of the usual His. Above pH 6, one of the two protons is lost. The remaining proton of the imidazole ring can reside on either nitrogen atom, giving rise to what are known as the N3-H or N1-H tautomers. In the N1-H tautomer, the NH group is nearer the backbone. Proofreading begins with separation of the mis-incorporated nucleotide from the DNA template. This pauses transcription. The polymerase then backtracks by one position and cleaves the dinucleotide that contains the mismatched nucleotide. In the RNA polymerase this occurs at the same active site used for polymerization and is therefore markedly different from the DNA polymerase where proofreading occurs at a distinct nuclease active site. The overall error rate is around 10−4 to 10−6.

=== Immobility === Disuse is a common cause of muscle atrophy and can be local (due to injury or casting) or general (bed-rest). The rate of muscle atrophy from disuse (10–42 days) is approximately 0.5–0.6% of total muscle mass per day although there is considerable variation between people. The elderly are the most vulnerable to dramatic muscle loss with immobility. Much of the established research has investigated prolonged disuse (>10 days), in which the muscle is compromised primarily by declines in muscle protein synthesis rates rather than changes in muscle protein breakdown. There is evidence to suggest that there may be more active protein breakdown during short term immobility (<10 days). Research has shown that immobilization induces anabolic resistance in skeletal muscle, reducing its responsiveness to anabolic stimuli such as insulin and β₂-adrenoceptor agonists like salbutamol.
