Pharmacologically the compound activates three receptors: GLP-1, GIP, and glucagon. GLP-1 and GIP signaling contribute to glucose-dependent insulin release, delayed gastric emptying, and reduced appetite, while glucagon receptor activation is associated with increased energy expenditure and hepatic fat oxidation. The single-molecule design is intended to keep these activities in one peptide rather than combining separate agents. Relative activity at each receptor differs, and the balance between them is a central question in interpretation. The glucagon component is partly offset by incretin-mediated insulin secretion, an interaction that remains incompletely characterized.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.

Quantification of the peptide in biological samples generally relies on liquid chromatography coupled with tandem mass spectrometry. This approach separates the analyte from matrix components and detects it by mass-to-charge transitions specific to the molecule. Immunoassays offer higher throughput but can cross-react with related peptides and metabolites, so mass spectrometric methods are preferred when structural confirmation is required. Method validation typically addresses accuracy, precision, selectivity, and stability under handling conditions.
Several questions remain unresolved. It is not yet known whether the compound reduces cardiovascular events or mortality, because outcome studies require long follow-up. The durability of weight reduction after treatment withdrawal is uncertain, and rebound has been observed with other incretin-based therapies. Long-term safety data covering several years are limited. Effects in adolescents, in pregnancy, and in people with significant kidney or liver impairment have not been characterized in published reports.
A 2005 epidemiology review concluded: "In aggregate the human epidemiological evidence is consistent with increased risk of birth defects in offspring of persons exposed to DU." Studies using cultured cells and laboratory rodents continue to suggest the possibility of leukemogenic, genetic, reproductive, and neurological effects from chronic exposure.
== Subfamily B2 == Subfamily B2 (also known as the adhesion G protein-coupled receptor family) contains receptors with long extracellular N-termini, such as the leukocyte cell-surface antigen CD97; calcium-independent receptors for latrotoxin and brain-specific angiogenesis inhibitor receptors amongst others. However, in some more recent classifications, they are no longer considered part of the same family as secretin (due to significant differences, like autocatalytic processing and different cell functions).

Two weeks after the band's formation, they were playing a gig at the University of Washington, trying to fill in a 40-minute set with a couple of original songs along with Hanoi Rocks and David Bowie covers. Diamond Lie gained attention in the Seattle area and eventually took the name of Staley's previous band, Alice N' Chains, then renamed Alice in Chains. Staley got permission from his former bandmates to use the name. Local promoter Randy Hauser became aware of Alice in Chains at a concert and offered to pay for demo recordings. However, one day before the band was due to record at the Music Bank studio in Washington, police shut down the studio during the biggest cannabis raid in the history of the state. The final demo, completed in 1988, was named The Treehouse Tapes and found its way to the music managers Kelly Curtis and Susan Silver, who also managed the Seattle-based band Soundgarden. Curtis and Silver passed the demo on to Columbia Records' A&R representative Nick Terzo, who set up an appointment with label president Don Ienner. Based on The Treehouse Tapes, Terzo signed Alice in Chains to Columbia in 1989. The band also recorded another untitled demo over a three-month period in 1989. This recording can be found on the bootleg release Sweet Alice.
=== Suggested limits on consumption of solanine === Toxicity typically occurs when people ingest potatoes containing high levels of solanine. The average consumption of potatoes in the U.S. is estimated to be about 167 g of potatoes per day per person. There is variation in glycoalkaloid levels in different types of potatoes, but potato farmers aim to keep solanine levels below 0.2 mg/g. Signs of solanine poisoning have been linked to eating potatoes with solanine concentrations of between 0.1 and 0.4 mg per gram of potato. The average potato has 0.075 mg solanine/g potato, which is equal to about 0.18 mg/kg based on average daily potato consumption. Calculations have shown that 2 to 5 mg/kg of body weight is the likely toxic dose of glycoalkaloids like solanine in humans, with 3 to 6 mg/kg constituting the fatal dose. Other studies have shown that symptoms of toxicity were observed with consumption of even 1 mg/kg.
Doris Ludlam, died 27 June 2002. Bridget Bourke, died 22 July 2002. Irene Crooks, died 20 October 2002. Ethel Hall, died 11 December 2002. All of these victims had been killed in Norris's first year of working as a nurse. Hall was a mother of one and a grandmother of two. Ludlam was a mother of two, a grandmother and great-grandmother who had worked as a nursery school teacher and fostered children for the charity Barnardo's. Wilby was a vulnerable widow. Crooks died on her 79th birthday, unable to ever open her cards or presents. Hall's son Stuart expressed his relief at Norris's conviction, stating: "I think he needs to be kept inside". He added: "He has got the knowledge to kill people and to do it discreetly. That makes him a danger to society and he must be kept inside. We hope Colin Norris never leaves prison and can never harm anyone else again." After the verdict was announced, Leeds Teaching Hospitals NHS Trust apologised to the victims' families for Norris's "disturbing" crimes, subsequently describing him as an "extremely dangerous criminal". In 2009, the Nursing and Midwifery Council struck off Norris from the medical register, taking just five minutes to come to a decision on the matter. Norris was imprisoned at HM Prison Frankland.

Immunodeficiency, also known as immunocompromise, is a state in which the immune system's ability to fight infectious diseases and cancer is compromised or entirely absent. Most cases are acquired ("secondary") due to extrinsic factors that affect the patient's immune system. Examples of these extrinsic factors include HIV infection and environmental factors, such as nutrition. Immunocompromisation may also be due to genetic diseases/flaws such as SCID. In clinical settings, immunosuppression by some drugs, such as steroids, can either be an adverse effect or the intended purpose of the treatment. Examples of such use include organ transplant surgery as an anti-rejection measure and in patients with an overactive immune system, such as in autoimmune diseases. Some people are born with intrinsic defects in their immune system, or primary immunodeficiency. A person who has an immunodeficiency of any kind is said to be immunocompromised. An immunocompromised individual may be particularly vulnerable to opportunistic infections, in addition to normal infections that could affect anyone. It also decreases cancer immunosurveillance, in which the immune system scans the body's cells and kills neoplastic ones. They are also more susceptible to infectious diseases owing to the reduced protection afforded by vaccines.
A documentary film (often called simply a documentary) is a nonfiction motion picture intended to "document reality, primarily for instruction, education or maintaining a historical record". The American author and media analyst Bill Nichols has characterized the documentary in terms of "a filmmaking practice, a cinematic tradition, and mode of audience reception [that remains] a practice without clear boundaries". Research into information gathering, as a behavior, and the sharing of knowledge, as a concept, has noted how documentary movies were preceded by the notable practice of documentary photography. This has involved the use of singular photographs to detail the complex attributes of historical events and continues to a certain degree to this day, with an example being the conflict-related photography achieved by popular figures such as Mathew Brady during the American Civil War. Documentary movies evolved from the creation of singular images in order to convey particular types of information in depth, using film as a medium. Early documentary films, originally called "actuality films", briefly lasted for one minute or less in most cases. While faithfully depicting true events, these releases possessed no narrative structure per se and were of limited interest. Over time, documentaries have evolved to become longer in length and to include more categories of information. Some examples are explicitly educational, while others serve as observational works; docufiction movies notably include aspects of dramatic storytelling that are clearly fictional.
== International == Chipotle had 17 locations outside of the United States by October 2014, with the majority in Canada, and the UK was in the process of opening more locations. The rate of overseas expansion was slower than expected. Many of the press reviewers thought that the food was overpriced for their area. In July 2023, the company announced it would partner with the Kuwait-based Alshaya Group in its first-ever franchise deal to expand into the Middle East. In April 2025, Chipotle announced it had signed a joint venture with Alsea to open restaurants in Mexico by early 2026, and possibly in other areas in which Alsea operates in Latin America. In September 2025, Chipotle announced it signed a joint venture with SPC Group to open restaurants in eastern Asia, with the first Chipotle restaurants opening in South Korea and Singapore in 2026. At the time of the announcement, Chipotle's existing international portfolio of restaurants included 60 locations in Canada, 20 in the United Kingdom, six in France, two in Germany, three in Kuwait and three in the UAE.
== Use == Dipipanone is now unavailable in most countries of the world either by laws prohibiting its medicinal use as in the United States or by falling out of production as more modern analgesics took its market share. Great Britain, Northern Ireland and South Africa are known to continue to use the substance but it is infrequently prescribed. Dipipanone is a Schedule I controlled substance in the United States; it has been assigned the ACSCN of 9622 and since 2013 had an annual manufacturing quota of 5 grams.

As of the mid-2020s retatrutide remains investigational and is not an approved medicine in the United States or the European Union. It has been supplied mainly to participants in clinical trials. Labels and availability can change, so regulatory listings should be checked directly.
Dual agonists act at the GLP-1 and GIP receptors, whereas retatrutide adds glucagon receptor activity on the same peptide. This third activity is the main structural and pharmacological distinction discussed in the literature. Direct head-to-head comparisons remain limited.
Reports usually give the study code LY3437943 alongside the name, the dose in milligrams, and the week at which an endpoint was measured. Percent body-weight change is the most frequent headline measure. Units and populations vary, so comparisons across reports require care.
